目的 研究鼠尾草酸(carnosic acid,CA)的遺傳毒性。方法 選用鼠傷寒沙門桿菌回復(fù)突變(Ames 試驗(yàn))、體內(nèi)哺乳動(dòng)物紅細(xì)胞微核、精子畸形以及單細(xì)胞凝膠電泳(彗星試驗(yàn))四項(xiàng)致突變生物學(xué)試驗(yàn)進(jìn)行篩選評(píng)價(jià)。結(jié)果 在6.25～50μg/mL 的劑量水平,CA 對(duì)鼠傷寒沙門菌株TA97、TA98、TA100、TA102 和TA1535 均無誘變性;此外,在受試劑量下小鼠骨髓嗜多染紅細(xì)胞微核、小鼠精子畸形以及體內(nèi)彗星試驗(yàn)的結(jié)果均為陰性(與溶劑對(duì)照比較,P> 0.05)。結(jié)論 在本實(shí)驗(yàn)條件下,CA 未見明顯遺傳毒性。

Objective To study the genotoxicity of carnosic acid (CA). Methods Salmonella Typhimurium reverse mutation assay (Ames test), in vivo mammalian erythrocyte micronucleus assay, sperm malformation assay, and single cell gel electrophoresis (Comet assay) were conducted to evaluate the genotoxicity. Results Under the concentration of 6.25—50 μg/mL, CA showed no mutagenic potency to the strains of TA97, TA98, TA100, TA102, and TA1535. Meanwhile in the test dose range, the genotoxicity indicated by mice bone marrow micronucleus assay, mice sperm malformation assay, and in vivo comet assay were negative (compared with solvent control, P>0. 0.05). Conclusion CA has no obvious genotoxicity under the test condition.
