目的 建立高通量檢測(cè)DPPH自由基清除能力的實(shí)驗(yàn)方法，對(duì)姜黃提取物抗氧化活性進(jìn)行初步評(píng)價(jià)。方法 通過測(cè)定抗壞血酸（維生素C）的DPPH自由基清除率曲線，以IC₅₀值作為評(píng)價(jià)試樣清除DPPH自由基能力的指標(biāo)，并將此應(yīng)用于測(cè)定姜黃醇提物和水提物清除DPPH自由基的能力。結(jié)果 測(cè)定波長(zhǎng)515 nm，抗壞血酸DPPH自由基清除能力在62.5～750.0 μg/mL線性良好，方法可靠。姜黃醇提物的DPPH自由基清除能力（IC₅₀ 7.78 mg/mL）明顯強(qiáng)于水提物（IC₅₀ 14.84 mg/mL）。結(jié)論 建立的DPPH自由基清除測(cè)定方法可靠、簡(jiǎn)便、靈敏，其高通量的快速檢測(cè)方法可為抗氧化藥物篩選提供參考。

Objective To establish a high-throughput method of DPPH free radical scavenging for evaluating the anti-oxidant activity of the extracts from Curcuma longa. Methods Taking IC₅₀ as the evaluating criterion of DPPH free radical scavenging capability by determining the scavenging rate curves of ascorbic acid, the DPPH free radical scavenging capability of two extracts from C. longa was determined. Results At the wavelength of 515 nm, DPPH free radical scavenging rate of ascorbic acid and concentration could fit a good linearity (0.062 5—0.75 mg/mL), and the results revealed that the method is convenient and reliable. Under this condition, the DPPH free radical scavenging capability of alcohol extracts (IC₅₀ 7.78 mg/mL) was significantly stronger than that of water extracts (IC₅₀ 14.84 mg/mL) from C. longa. Conclusion The convenient and reliable method could be used to evaluate the anti-oxidant activity of other Chinese materia medica, as well as provide an approach and reference for high-throughput activity screening.
