目的 探討桂枝茯苓膠囊對子宮肌瘤大鼠的治療作用及其機制.方法 采用注射苯甲酸雌二醇和黃體酮復制大鼠子宮肌瘤模型,造模大鼠按體質(zhì)量隨機分為模型組、他莫昔芬(枸櫞酸他莫昔芬片3.6 mg/kg)組以及桂枝茯苓膠囊0.14、0.27、0.54 g/kg組,另設(shè)對照組.連續(xù)給藥8周后處死大鼠,取子宮、脾臟、胸腺并計算其臟器系數(shù),比較子宮病理評分和組織形態(tài),放射免疫分析方法檢測血清中雌二醇(E2)、孕酮(P)及子宮組織中表皮生長因子(EGF)水平.結(jié)果 桂枝茯苓膠囊顯著降低大鼠子宮系數(shù),減輕子宮壁平滑肌增生,顯著降低血清E2和子宮組織EGF水平.結(jié)論 桂枝茯苓膠囊對大鼠子宮肌瘤具有治療作用,其作用機制可能與降低血清E2、子宮組織EGF水平有關(guān).

[Abstract]
Objective To discuss the therapeutical effect of Guizhi Fuling Capsule on uterine leiomyoma in rats and its mechanism. Methods Uterine leiomyoma model in rats was made by injection estradiol benzoate and progesterone. The rats with uterine leiomyoma were randomly divided into model group, tamoxifen (Tamoxifen Citrate Tablets 3.6 mg/kg) group, and Guizhi Fuling Capsule (0.14, 0.27, and 0.54 g/kg) groups. There was also a control group. After treatment for eight weeks, the rats were killed by cervical dislocation. The organ coefficients of the uterus, thymus, and spleen were calculated. Pathological score and pathological changes of uterus tissue were detected. The levels of estradiol (E2), progesterone (P) in serum and epidermal growth factor (EGF) in uterus tissue serum were determined by radioimmunoassay. Results After treatment, Guizhi Fuling Capsule significantly reduced the uterus coefficient, alleviated hyperplasia of smooth muscle in uterus wall, significantly reduced the levels of E2 in serum and EGF in uterine tissue of rats. Conclusion Guizhi Fuling Capsule has therapeutical effect on uterine leiomyoma in rat, and the mechanism may be associated with reduction of E2 levels in serum and EGF levels in uterine tissue.
