目的 對(duì)產(chǎn)于我國(guó)甘肅省的中藥秦艽的3種基源植物秦艽、麻花秦艽與小秦艽進(jìn)行RAPD分析,建立具有鑒別意義的DNA指紋圖譜.方法 采用RAPD分子標(biāo)記對(duì)秦艽的基因組DNA進(jìn)行PCR擴(kuò)增,利用NTSYSpc 2.1和Popgene 3.2軟件分析3種秦艽間的親緣關(guān)系,構(gòu)建樹系圖.結(jié)果 從80條RAPD引物中篩選出4條具有鑒別意義的多態(tài)性引物,在3種秦艽中共得到39個(gè)DNA條帶,其中共有條帶2條,多態(tài)條帶37條.據(jù)此獲得了能有效區(qū)別3種秦艽的多態(tài)性RAPD指紋譜.結(jié)論 甘肅產(chǎn)秦艽的3種基源植物遺傳分化明顯,RAPD方法能有效地將它們區(qū)分鑒別.
